Purification

Choose the desired Purification. If no purification is chosen Oligo will be delivered in standard LowSalt condition.

Cartridge:
Cartridge purification more efficiently removes leftover salts from the (post)synthesis process and short sequences. Cartridge purification is not recommended for oligos longer than  50 bases.

HPLC
HPLC purification removes shorter sequences from the full length product as well oligos that are not modified when a modification is ordered.
Reverse-phase HPLC is usually not recommended and performed for the purification of oligos longer than 50 bases.
Twice Dyed Probe, XS probes and Molecular Beacon are always HPLC purified.

PAGE:
PAGE is the recommended technique for purifying oligos >50 bases long. Yields from PAGE are lower than from other methods due to the relative inefficient extraction of oligos from the gel.
For very long Oligos 2xPAGE might be advisable, please consider the further yield reduction.

Lyophilized

Oligos in tubes are standardly shipped dissolved as 100µM TE if not other mentioned. This is the most stable storage method.

Biolegio Guidelines and Orderform Manual

To avoid that your ordered oligos won't meet the expected specifications, there are a few guidelines.

The fields in orange are mandatory and will appear on the order conformation and shipment documents.

Comment

It is possible to fill in any comment regarding the oligos. The first 20 caracters of the comments will be printed on the tube labels.

Researcher name

It is possible to fill in the name of the researcher name. The first 20 caracters of the name will also be printed on the tube labels.

Oligo Name

Fill in the name of the Oligo. Max 25 letters will be printed on the oligo tube.

 

Oligo Sequence

Fill in the oligo sequence:

·         Internal modifications are labeled as "1" and "2". If more than two different internal modifications are to be implemented please contact Biolegio.

·         Inosine must be entered with the letter " I ". Uridine must be entered with the letter " U ".

·         The standard codes for wobbles (degenerated bases) are: R=A/G, Y=C/T, M=A/C, K=G/T, S=C/G, W=A/T, B=C/G/T, D=A/G/T, H=A/C/T, V=A/C/G, and N=A/C/G/T.

·         Please avoid special characters in the "sequence" fields.

Scale

Please make sure to enter the synthesis scale. Otherwise your oligos will be synthesized by default on the 40 nmol scale.
Fill in the desired synthesis scale. Oligos shorter than 11 or longer than 80 bases will be charged at higher prices, please enquire.

·         10 nmol scale: up to 40 bases

·         40 nmol scale: up to 200 bases **

·         200 nmol scale: up to 300 bases **

·         1000 nmol scale: up to 300 bases**

·         Minimum scale for Twice Dyed Probes, XS probes, Molecular Beacons or modified oligo's is 40 nmol

·         Minimum scale for purified oligos is 40 nmol scale

·         Minimum Scale for NGS-Standard, NGS-Stand Plus and NGS-Premium is 40 nmol

 

(** For long oligos we recommend additional purification. The purification depends on the application the oligos are used for. More info in Section purification)

All NGS-Oligos are of highest quality. To avoid cross contamination we resign to use column (Cartridge or HPLC) purification for our NGS-Oligos which are generally used for multiple Oligos.

Please choose between:

NGS-Standard: Highest quality oligos with minimized cross contamination desalt.
NGS-Standard Plus: Highest quality oligos with minimized cross contamination, PAGE purified for highest purity and LC-MS checked.
NGS-Premium: Ensure highest yields and quality. NGS-Premium Oligos comes with guaranteed amounts in the chosen scales for unmodified Oligos up to 80 bases.
NGS-Premium Oligos are PAGE purified, LC-MS checked and LC-MS documentation is provided.
       40 nmol =   10-   20 nmol yield
     200 nmol =   40-   50 nmol yield
   1000 nmol = 100-150 nmol yield
